Los Angeles: A breakthrough by US scientists has brought the prospect of an anti-agieng pill closer.

The team, led by Dr Valter Longo, a biomedical gerontologist at the University of Southern California, have successfully prolonged the life of yeast cells by up to six times its usual lifespan.

Dr Longo said the next step would be to exploit this understanding to create a drug that could reverse human ageing.

The research, in which scientists removed two key genes – SCH9 and Sir2, forced the single celled organisms into “survival mode.” SCH9 governs the abiity of the cell to convert nutrients into energy, while Sir2 restricts the lifespan by converting energy into growth and reproduction. The removal of the genes tricked the cells into believing they had no food and so they concentrated on survival and lived for six weeks instead of one.

The experiment also confirms earlier research in which flies, worms and mice, who were put on a calorie restricted diet lived longer.

Stress may help fight breast cancer: new research in Denmark

Copenhagen: High levels of daily stress appear to result in a lower risk of developing breast cancer for the first time, says a study in this week’s British Medical Journal.

But high stress may put women at risk of other serious illnesses warn the researchers, a team from Denmark.

The findings follow an eighteen year study of over 6,500 women in Copenhagen. At the start of the study researchers asked the women what levels of stress they experienced routinely in their lives, and classified the results into low, medium and high levels. Stress was defined as tension, nervousness, impatience, anxiety, or sleeplessness. (Stress levels were not measured throughout the study.)

In calculating the effects of stress, researchers also adjusted the results for other factors, such as whether they had children or whether they were menopausal, which would have an influence on developing breast cancer. They did not account for risk factors such as family history of the disease however.

Of the 251 women diagnosed with first-time breast cancer over the study period, researchers found that women reporting high levels of stress were 40% less likely to develop breast cancer than women reporting low levels of stress.

The study further found that, for every increased level of stress on a six-level scale, women were 8% less likely to develop breast cancer.

One explanation for the findings may be that sustained levels of high stress may affect oestrogen levels – which, over time, may have an influence on developing breast cancer. But this theory has not been tested, and research in this area so far has mainly been restricted to animals, caution the authors.

Despite the findings, the authors warn that stress-induced changes in hormonal balances are not a healthy response, and continued stress may play a damaging part in other illnesses – particularly heart disease.

Secret of olive oil is anti-ageing

Rome: New research has pinned down why extra-virgin olive oil, a staple of Italian cuisine, helps the nation’s inhabitants avoid tumours and other diseases in their later years.

The results of a nine-year study of Italians living in the olive-rich southern region of Puglia showed recently that they were statistically less prone to cancer and other ailments because they consumed the oil all their lives .

Now researchers at the Monell Chemical Senses Center in Philadelphia have discovered precisely why this is the case .

They have identified in extra-virgin olive oil a molecule which is similar to ibuprofen, an anti-inflammatory agent contained in several over-the-counter painkillers .

They believe that the molecule, which they have called ‘oleocantale’, is the key element which gives olive oil its legendary properties .

If assimilated over a long period of time, it can mean a person is protected from tumours and other problems that regularly crop up when a person becomes old, they say .

The research, which finally offers scientific support for what doctors and nutritionists have said for years, appears in the September edition of the journal Nature .

The discovery comes just five months after scientists at Bari University’s Geriatrics Department offered convincing statistical evidence that olive oil is a real ‘elixir of life’ .

Unveiling a nine-year study of over 700 people in Puglia aged 65-84, they said extra-virgin olive oil clearly has a string of health benefits such as its ability to combat chronic diseases and, above all, guard against tumours .

The survey aimed to assess the role of diet, and in particular intake of monosaturated and polysaturated fatty acids, on ageing and death .

The 704 Puglians ate a typical Mediterranean diet in which fat (17.6% monosaturated fatty acids, 3% polysaturated and 8.4% saturated) accounted for 29% of total energy intake .

Extra-virgin olive oil provided 85% of the monosaturated fatty acids .

The latter were associated with reduced mortality, for all causes, the study said. In particular, 15 grammes a day of monosaturated fatty acids cut deaths among over-65s by a fifth .

“This is proof that not only is olive oil a healthy food product but a splash of it a day helps prevent tumours,” said Professor Giorgio Calabrese of the National Authority for Food Safety .

An Athens University study also showed recently that the mortality rate among the elderly was significantly lower in Mediterranean countries like Greece, Spain and Italy .

Nutritionist Antonio Migliaccio commented: “Extra-virgin olive oil has great anti-oxidant powers and is therefore recommended in low-calory diets. It also increases so-called good cholesterol and lowers bad cholesterol.”

New gene plays role in ageing, scientists discover

Texas: Scientists in the United States have discovered a gene that can keep mice alive for 30% longer than normal.

They say the gene, called Klotho, has a key role to play in many of the processes related to ageing. Mice and people with defective forms of the gene appear to age faster.

Because humans have a very similar version of the gene, the hope is that it will show a way to improve our declining years.

Now researchers have shown that by boosting the activity of the gene, they can extend the natural lives of male mice from two to three years. The effect is not quite so strong in female mice.

“It could be one of the significant steps for developing anti-ageing therapy,” Dr Makoto Kuro-o, assistant professor of pathology at the University of Texas’ Southwestern Medical Center and senior author of the study, told Science magazine.

Klotho seems to delay many of the effects of old age, like the weakening of bones, clogging of the arteries and loss of muscle fitness.

This is important for those researching the causes of ageing, whose intention is not so much to prolong life as to improve the quality of our final years.

But there may be downsides with Klotho. The long-lived mice in the new experiments tend to be less fertile. And the gene may also predispose people to diabetes.

The trick for researchers will be to find ways of getting the life-enhancing results of Klotho while avoiding the drawbacks. Klotho seems to delay the effects of old age in mice.

New York: Emma Samms, the former Dynasty actress, is the glamorous face of Isolagen, a unique skin rejvenation system, that harnesses a person’s own cells and grows them to rejuvenate wrinkles.

This is the Isolagen process in detail:

Isolagen’s patented autologous (a patient’s own cells) living cell therapy, or ACS, begins with the injection of a local anesthetic to numb a small section of skin behind the ear. This area was chosen because of its vascularity, lack of sun exposure, and the invisibility of any scarring.

A simple punch biopsy is used to obtain a small 3 mm piece of skin tissue, which is packed in an appropriate container and shipped overnight to the Isolagen laboratories. The minimal incision is closed with an adhesive or single absorbable suture.

At Isolagen, the tissue is cultured utilizing Isolagen’s patented ACS process. This process separates collagen-producing cells, called fibroblasts, from the rest of the tissue then stimulates them to multiply into tens of millions of new cells.

After approximately six weeks, 1 to 1.5 ml of cultured fibroblasts are sent back to the doctor for injection into the patient’s wrinkles, lines, and scars.

Generally three sets of injections will be performed, about two weeks apart, with tens of millions of collagen-producing cells being injected during each visit.

Within the patient’s skin, it is believed that the injected fibroblasts will continue to multiply and create new collagen that may fill dermal imperfections and wrinkles, and may reduce the signs of aging.

Cryogenic storage of cultured cells may also permit patients to receive future treatments with cells that were harvested when the patient was younger. It is unknown at this time whether this would represent a benefit to patients.

Since the Isolagen Process is an autologous system (exclusively using a patient’s own cells), it is anticipated that there may be a substantially reduced possibility of allergic reaction as compared to bovine collagen and other non-natural fillers.

Isolagen hopes to demonstrate that the use of autologous cells will result in prolonged beneficial effects, as the immune system should not reabsorb or reject them as it might with foreign materials and proteins. Patients may experience gradual and continued improvement as a result of the natural activity of the re-introduced cell structure.

Anti-ageing skincare for men launches

Nevada: Bio-Vel Technologies says it has launched the first anti-aging skin care system designed and marketed exclusively for men onto the US market. The range includes a number of innovative ingredients aimed at tapping into the growing market for increasingly sophisticated male cosmetic and personal care products.

The Elements for Men range contains two of the newest designer peptides, manufactured by Spanish pharmaceutical innovator Lipotech, as well as one of the oldest and time proven skin protectants known to man – shark liver oil.
“We originally went into business with the idea of developing a new anti-aging skin care line for women,” says Bio-Vel CEO Gina Gaede. “But when we saw how the market was completely inundated with women’s products and how the expanding male market was virtually untapped, we re-focused our sights on developing a line of products exclusively for men.”

CoEnzyme Q10

CoEnzyme Q10 is a fat-soluble vitamin-like substance present in every cell of the body. It also occurs naturally in a number of foods, including organ meats, soy oil, sardines, mackerel and peanuts. It is a powerful antioxidant and acts as a coenzyme for several of the key steps in the production of energy within every cell. Low levels of CoQ10 have been linked to several deseases. ESSENTIAL FATTY ACIDS

EFAs are essential to the body and is able to manufacture them aside from two – linoleic acid (omega-6 family) and linolenic acid (omega3 family). Since they must come from the diet they are classified as EFAs.

Omega 3 fatty acids are found in cold-water fish, perilla and flaxseed oils. They contain a substance known as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). These are usually lacking in the Western diet which has mainly omega-6 fats supplied in the diet from meat and vegetable oils.

Gamma linolenic acid (GLA) which is found in evening primrose oil, borage oil and blackcurrent oil is an important fatty acid that plays a beneficial role in prostaglandin formation

EFAs interfere with blood clotting so anyone contemplating surgery or suffering from a hemorrhagic disease should consult a doctor before taking them.

DHEA (dehydroepiandrosterone)

The supplement DHEA, is a steroid hormone that can slow ageing, prevent disease and improve strength. Its medical uses include the treatment of depression and dementia It is used by some athletes to increase muscle mass and strength, but banned by sporting bodies including the International Olympic Committee and the National Football League. It is classed as a food supplement and has also become popular as an anti-ageing remedy.

Ageing men and woman are invariably DHEA deficient. DHEA is the most abundent circulating hormone in the youthful human body but peaks by the age of 25 and declines thereafter. By the age of 70 most people DHEA has declined by over 80%. DHEA should not be taken by those with existing prostate or breast cancer. Those with liver disease should take dissolve in the mouth tables rather than capsules.

DHEA levels decrease after 30 and it is thought that some symptoms of ageing could be associated with a DHEA deficiency.There has been considerable interest in using DHEA in Alzheimer’s disease. DHEA supplements in menapausal woman have also shown to increase estrogen levels and reduction in hot flushes.

A study by the Univesity of California, San Diego, said that 50mgs a day taken daily over six months increased lean body mass and muscle strength.

Known side effects: DHEA and pregnenolone may cause heart rhythm irregularities and irreversible hair loss.

DMAE

DMAE, or dimethylaminoethanol (chemical name deanol), is a compound found in high levels in oily fish such as anchovies, sardines and salmon. It is also produced in small amounts by the human brain. It is sold as a food supplement to boost brain function and is thought to be helpful in attention deficit hyperactivity disorder (ADHD). Doctors are also looking at its role in treating poor memory and Alzheimer’s Disease, as well as some movement disorders.

The supplement was originally researched as a treatment for ADHD and early studies in the 1970s revealed that it was helpful for children with learning disabilities and behavior problems. This substance appears to increase production of chemicals in the brain essential for short-term memory, concentration, and learning capacity. It is also classed as an “cholinergic” because it is thought to increase levels of the neurotransmitter acetylcholine, one of the chemicals in the brain that enhances mental powers. A decrease in the brain cells that produce acetylcholine is thought to be parlty responsible for Alezheimer’s. “Cholinergic” drugs, such as tacrine (Cognex), donepezil (Aricept), rivasatigmine (Exelon), and galantamine (Reminyl) are used to treat the dementia of Alzheimer’s disease.

Although there are no conclusive studies it is possible that DMAE has memory-boosting effects. Many nutritionally oriented physicians prescribe DMAE along with another memory enhancer, the dietary supplement phosphatidylcholine.

DMAE supplements won’t work for everybody and are not intended as a cure. But they are safe and may be helpful. They should be taken with meals.

There are no known drug or nutrient interactions associated with DMAE but more needs to be learned this supplement. You should consult a doctor, particularly if you have any illnesses such as epilepsy.

International anti-ageing skin doctor Nicholas Perricone describes DMAE as “the magic bullet” for skin. He prescribes supplements and a diet high in wild salmon – his research documents changes in the skin in only three days. GLUCOSOMINE

Glucosamine is a natural substance in the body known as an amino-sugar which is made in the body as a result of the synthesis of L-glutamine and glucose. Glucosamine stimulates glycosamino-glycans which are important in the formation of cartiliage and joint ligaments. As people age, they lose the ability to manufacture sufficient levels of glucosamine. Cartilage then loses its gel-like nature and ability to act as a shock absorber. Research in The Lancet medical journal which used X-rays to investigate the knee cartilage of volunteers, found those who took glucosamine pills for three years halted the progress of their osteoarthritis. In Spain, Portugal and Italy, glucosamine is the preferred treatment for arthritis. Glucasamine is sold as a supplement and there are also skin patches. It is often sold in supplements with Chondroitin sulfates which help the glucosamine into joints.

HUMAN GROWTH HORMON

HGH is released by the pituitary gland during sleep and is the most important anti-ageing hormone of all. It is produced until around mid-20s to age 30, but continues to decrease as we age. Genuine HGH is synethically made by only two drugs companies mainly for use with children who suffer from growth hormone deficiency. In adults HGH injections are used to increase metabolism and other visible signs of ageing but these are only available on prescrption. Naturapathic HGH, which is widely available on the web, has been criticised as a “scam” by professional anti-ageing experts.

HGH restores muscle strength, stamina and other attributes of youth.
There are also natural ways to encourage HRH production, including exercise. Humans could live 5000 years say scientists

Blame it on boomers – people born between 1946 and 1965. The baby boom generation now makes up more than a quarter of the US population population -some 77.5 million people, with more than 160,000 in San Diego. One-third are over age 50.

Every seven seconds, another boomer joins that group. In just seven years, the first boomers will hit official retirement age. By 2030, boomers 65 and older will represent one in every five people.

For them, 65 will be the new 45. Or so they hope, and so many claim.

From self-described anti-aging institutes to miracle elixirs to how-to manuals for living a century or more, boomers (and just about everyone else) want to live longer than those in previous generations.

These days, the average American has a life expectancy of 76.9 years — a little more for females, a little less for males.

Most people, of course, want to live much longer than the average. But what are the odds of living to the century mark and beyond? And how much beyond is possible?

The good news is that most scientists think human life expectancies can be substantially stretched. The not-so-good news, some experts say, is that the estimated maximum human life span — about 125 years — seems fairly fixed and that most of us will never get close.

“Longevity is really a modern phenomenon,” said S. Jay Olshansky, a demographer and biologist at the University of Illinois at Chicago. “The vast majority of humans ever born died before the age of 10, usually from infectious diseases.

“We’ve done fabulous things to boost the survival rates of the young — improved sanitation, new medicines — but now it’s a whole different ballgame. It’s not so easy to add 70 years of life to somebody who’s already 70 years old.”

No doubt. But a number of scientists and doctors think it’s too early to start talking about a “finished” line. They assert, in principle, that there is no maximum human life span.

Aubrey de Grey, a biogerontologist at the University of Cambridge in England, says that under the right circumstances, humans born in the 22nd century (just 96 years away) could live up to 5,000 years.

De Grey, who advocates using technology to develop a “true cure for aging,” is indisputably at the optimistic extreme. But plenty of others see longer lives ahead.

“I think people will someday live substantially longer than today,” said Steven Austad, a biologist at the University of Texas Health Sciences Center in San Antonio. “(Living) into your 100s will be fairly routine, up to 150 for the outlier (a longer-lived person who is the exception to the rule). I think this because we have been so successful at figuring out how to make animals live longer.

“The arguments (against appreciably longer life spans),” he added, “are based so far as I can tell on ignoring a huge pile of research done over the past 15 years and the mystical belief that longevity, unlike every other human trait we know of, is impossible to change.”

Wear and tear

In biological terms, aging is usually defined as the accumulation of random damage to the building blocks of life, most notably DNA. The damage starts in early childhood and accelerates after age 30 or 40.

Over time, the accrued, unfixed damage impairs bodily functions. Cells, tissues, organs and systems work less well.

The immune system of a typical 65-year-old, for example, is only one-tenth as effective as that of a teenager. With less protection comes greater vulnerability to disease.

“Aging, in our view, makes us ever more susceptible to such ills as heart disease, Alzheimer’s disease, stroke and cancer,” Olshansky wrote, along with Leonard Hayflick, a gerontologist at the University of California San Francisco, and Bruce A. Carnes, a professor of geriatric medicine at the University of Oklahoma Health Sciences Center, in an online essay published this year by Scientific American magazine.

But conditions like heart disease are age-related, not the actual equivalent of aging, the scientists note. And aging is not the same as longevity.

Even if modern medicine could eradicate all the leading causes of death among the elderly, says Hayflick, an early pioneer in gerontology research, human life expectancy would increase no more than 15 years.

People would still age, he said. Other afflictions would rise up to exact their deadly toll. The maximum human life span would remain unchanged.

Age-old questions

If scientists want to boost that maximum life expectancy of about 125 years, most experts say, they’ve first got to solve the questions of how we age and why.

All organisms age, but the process, called senescence, is variable and, in some species such as the giant tortoise and rougheye rockfish, it’s virtually negligible.

The tortoise is known to live for 150 years or more; the rockfish more than two centuries. Both exhibit almost no signs of aging.

Variable senescence among species suggests to researchers that there are biological mechanisms, as yet undiscovered or understood, that might be altered, replaced or removed to effectively slow or even reverse aging in humans.

De Grey at the University of Cambridge says biotechnology is the answer.

He thinks current and foreseeable medical technologies, from drugs that repair or prevent cellular damage to organ regeneration and replacement, may soon be able to reverse the effects of aging. He predicts that researchers will actually do so in mice during this decade.

“Intervention to remove the accumulating damage . . . has the potential to postpone aging indefinitely,” he said.

A big key will be genes, which researchers say dictate and exert influence over roughly 30 percent of the aging process.

“As we begin to learn more about genetics, we see that there perhaps are certain genes that enable people to cope better with stress, react better to hormones and possibly regulate the rate of aging,” said Dr. Robert Butler, president of the International Longevity Center-USA, a New York City-based think tank.

Scientists are pushing hard to find such genes.

In 2001, Harvard University physicians and molecular biologists conducted tests on people who were all at least 90 years old and found they shared one or two genes on a specific chromosome. The exact function of these genes, however, has not been determined.

More recently, Olshansky and colleagues have launched a global project to identify so-called longevity genes by sampling DNA from exceptionally elderly people in places where very long life spans are common, such as Okinawa, Japan; the Vilcabamba valley in Ecuador; and the Hunza region of Pakistan.

Such efforts, though, won’t mean more birthdays for everyone anytime soon.

Most researchers are skeptical that there will ever be a one- stop genetic remedy. They note that aging involves lots of other factors, not to mention the considerable ethical and social issues attached to significantly modifying the human genome.

“Is the purpose of medicine and biotechnology, in principle, to let us live endless, painless lives of perfect bliss?,” the President’s Council on Bioethics asked in a report last year. “Or is their purpose rather to let us live out the humanly full span of life within the edifying limits and constraints of humanity’s grasp and power?”

The council expressed concern that a world full of centenarians and a diminished sense of mortality might result in problems no one can imagine or resolve.

Others suggest that such worries miss a more pertinent point.

Tom Perls, a geriatrician who runs the New England Centenarian Study, says most people are already genetically well-equipped to live reasonably healthy lives well into their 80s. The only requirement: They take good care of themselves.

Dr. Dilip V. Jeste, director of the Stein Institute for Research on Aging at UCSD, agrees.

“The obstacles over which we have control (of aging) are primarily environmental and behavioral,” Jeste said. “These include smoking, use of drugs of abuse as well as excessive alcohol, sedentary habits, poor nutrition, etc.

“The prevention of hypertension, diabetes and obesity may help increase life span significantly,” he said. “Resilience, optimism, adaptation to changing circumstances and optimal coping style are also important.”

Jeste said the focus of aging science should be less about extending life spans and more about making aging a “successful process associated not only with longevity, but also with a high level of activity of brain and mind.”

The record for the longest documented life is held by Jean Calment, who died in France in 1997. She was 122. The last years, however, were not kind. She was blind, deaf, incontinent and unable to care for herself.

Quantity wasn’t quality, and Calment most likely longed for the youth of her 90s.

Hormone Replacement Therapy

In anti-ageing hormone replacement therapy, a qualified doctor will take detailed blood/saliva/urine tests to determine current levels of hormones in the body. As we age levels of most hormones deteriorate and contribute to physical and mental signs of ageing including weight gain and memory loss.

These are just some of the hormones that might be supplemented as part of an anti-ageing programme:

Human Growth Hormone (Somatotrophin)

It is manufactured in the laboratory and until 1996 was only authorised by the US’s FDA in the treatment of children with HGH deficiencies which led to abnormal height.

HGH can still only be administered on prescription but it is approved for use in hormone-deficient adults. This hormone decreases with age and can improve cognitive function including memory, muscle mass, decrease in body fat, healthier heart, lungs, kidneys, increased bone density, elevation of mood, energy and libido.

There are also some unacceptable side effects such as a possible role in promoting the growth of cancer and its use is still experimental as not enough is known. There are a number of web sites selling “homeopathic” HGH but experts say only injectable HGH works because it by-passes the stomach where it would loose its properties.

When doctors administer HGH the therapeutic levels are monitored by the levels of another hormone called Insulin Growth Factor (IGF-1) as HGH can only be detected in the blood for onlya few minutes after it is administered. The amount of IGF-1 in the blood increases as we get older and also in cancer patients and there is a strong link between IGF-1 and cancer. The transdermal administration of HGH lowers this risk.

This therapy is available from anti-ageing doctors on this site. You are advised to seek a blood test prior to treatment to see whether you are deficient in this hormone in the first instance.

The Baxamed Medical Center has a “Youth Restoration Programme” in which a blood analysis determines the level of hormones such as HGH, Testosterone, Estrogen, Progesterone and DHEA, of which decline with age. At the clinic they replace the missing hormones in the correct dosage. The clinic also offers fetal cell therapy which is injected intramuscularly. The patient then continues the treatment at home.

DHEA – Dehydroepiandroserone

DHEA is the most prolific steroid hormone produced by the human body but it declines with age. aIt helps build muscle, loose fat, normalize cholesterol and even improves memory. DHEA levels decline quite rapidly with advanced age.

MELATONIN

Melatonin is secreted by the pineal gland located near the bottom of the brain and regulates the body’s sleeping pattern. Again as we grow older, the pinal gland calcifies and the secretion of Melatonin declines, causing the sleeplessness so often suffered by the elderly.

Melatonin also helps with stress and helps the immune system repair itself. It is helpful for migraine and has been shown to slow down the growth of cancerous cells in certain types of cancer. Its role in anti-ageing is that melatonin is the regulator of other hormones released from the brain and diminised secretation may be responsible for ageing.

PREGNENOLONE

Pregnenolone like DHEA, Melatonin, Estrogen, Progesterone, Testosterone and Human Growth Hormone is a hormone that declines with increasing age. Recent studies demonstrated that Pregnenolone has a remarkable capability to increase memory function at very low doses. It is used to treat memory impairment due to illness and old age. In the 1930 to 1940s it was successfully used to treat arthritis, but it was forgotten when the first anti inflammatory medications were pushed on the market.

TESTOSTERONE

What is the Andropause: women in their fifties enter menopause, caused by a rapid decline of sex hormones – particularly progesterone. Until recently it was only suspected that men go through a change like menopause. New evidence confirms this supspicion. The decline of the male sex hormone testosterone is much slower than in women making the symptoms develop more slowly and hardly noticeable.

Testosterone is responsible for libido and sperm production. But it is not only a sex hormone, it also increases protein synthesis in the cells, and keeps up aerobic metabolism thus reducing the risk of cancer as cancer cells use anaerobic metabolism. Aerobic metabolism is also important to regulate cholesterol, supply sufficient oxygen to vital organs like the heart and the pancreas thus helping to prevent heart disease and diabetes. At any age aerobic exercise can increase the production of testosterone. Testosterone has fibrinolytic properties, this means it helps to prevent blood clots that would lead to thrombosis. Testosterone has anabolic properties and is counteracted by cortisol and adrenalin. Constant stress increases cortisol and adrenalin and aging decreases testosterone secretion. This leads to a catabolic state, where muscle mass is lost.

Testosterone therapy should be done under the supervision of a specialized physician. Only natural testosterone should be used such as Testosterone cream or patches. Most other forms are synthetic and not identical to the bodys’ own testosterone.
